Immature embryos of corn (Zea mays L.) with 8 genotypes were cultured on the media containing 2 mg/l 2,4-D and 20 mM.L-proline. Regeneration of plantlets from embryogenic callus was achieved when genotypes Ki1, Ki3, SW2 and SW3 were cultured. Sixty-five genotypes had been studied for anther culture using 15-different recipe media. Only 2 genotypes, SW3 and CH13 produced callus on Y3 and Y31 media respectively. Callus lastly died without regeneration into plantlet. Protoplast isolation has been done on SW1 and SW3 with various enzyme combination used. 2 % cellulase added with 0.2 % macerozyme yielded more protoplasts than 1 % cellulase. The activity of chitinase enzyme had been checked among 20 genotypes. Inbred line Ki21 showed the maximum quantity of chitinase enzyme extracted from kernels with 2.86 Unit/g seed.
