The nitrogen status of the medium during micropropagation

During the micropropagation the plants are grown on artificial medium. The plants take up the elements which are important for their growth from the medium. The question is, whether the N content of the medium is taken up and how it is utilised by the plants under the effect of different types of illumination and photoperiods. In the experiments in vitro cultured J-TE-F apple rootstocks were used. The plants were grown on liquid Murashige-Skoog medium for 34 days. The change of N content of the medium was observed with help of Contiflo automatic analyser. It was determined that the N content of the medium decreases till the 22nd and 26th day (depending on the treatments). After this time on the 34th day N content of the medium increases and finally decreases to zero independently from treatment. It was established furthermore that the procedure and sterilisation of medium doesn't influence the available N content of the medium
