Tissue culture of sugarcane genotypes

Tissue culture of 10 sugarcane (Saccharum officinarum) cultivars, Coj-82, Coj-83, Coj-84, Co-1148, CP72-2086, SPSG-232, CP77-400, SPSG-160, CP 1210 and CPF-224 were studied using Modified MS Medium with Pith parenchyma tissue and cut edges of leaves were used for callus culture with five levels (1 mg/1, 3mg/1, 5ml/1, 7mg/1 and 9mg/1) of 2, 4-D in the medium. Response to initiation of callus was different in various genotypes, 2, 4-D concentrations and explant sources. In general callus initiated within three weeks of explantation. Genotype SPF-232 was top callus producer and CPF-224 was at the bottom. Lower levels of 2, 4-D were better and six out of ten varieties produced maximum callus at 1mg/1 of 2, 4-D. Leaf as explant source was better for callus production and shoot initiation from callus. Similarly leaf callus derived from lower 2, 4-D level performed better in shoot initiation than the pith callus. Shoots on transfer to 1/2 MS medium devoid of growth regulators induced rooting. Regeneration of shoots in various genotypes varied from 38.8 to 2.35 shoots per test tube. Callus production did not show any direct relation to plant regeneration.
