Population structure analysis of Pseudomonas solanacearum causing bacterial wilt in tomato using RAPD-PCR

Genomic DNA of the different biovars of Pseudomonas solanacearum isolated from tomato were extracted using modified CTAB method. The concentration of the DNA recovered ranges from 100-500 mg/ml which was estimated by comparing the intensity of banding with the known concentration of indigested DNA. Twenty OPERON Kit D RAPD primers were applied to the P. solancerum genomic DN for PCR reaction. The generated DNA fragments were scored and analyzed using Numeric Taxonomy System (NTSYS) Program. Several OPD RAPD primers viz OPD-18, OPD-11, OPD-01 gave the best banding profile to analyze the similarity and differences of the isolates among and within biovars of P. solanacearum. Other primers gave very few to no banding
