Optimization of conditions for the extraction of mitochondrial DNA of Oreochromis sp.

Mitochondrial DNA of Oreochromis sp. was extracted using four protocols: a modified Alkaline-Detergent method (A-D) (Knox and Verspoor, 1991), and three different phenol extraction methods by (1.) Villasenor et.al. and Chapman and Powers (1990) (PE-1), (2.) Bernatchez et. al. (1988) (PE-2), and (3.) an integration and modification of (1) and (2) (PE-3). Liver, eggs, ovary, testis, spleen, and blood were used and their efficiency as sources of mtDNA was determined based on quantitative and qualitative analysis. Quality was checked by 1 percent agarose gel electrophoresis as well as by UV spectrophotometry. The molecules weight of the mtDNA was found to be approximately greater than 23 kilobases, but the probability of heterogenity existing within species exists. Statistical analyses using Analysis of Variances and Duncan's Multiple Range Test were done to compare mtDNA concentration and purity obtained from the different protocols and tissue sources at a 95 percent confidence level
