Stimulating lysine excretion by using polymixin-B sulphate

Product accumulation in the medium can be enhanced either by stimulating excretion system or weakening uptake systems. In the present study, the effect of several antimicrobial agents on lysine fermentation using a strain of Brevibacterium lactofermentum was observed. The agents selected were penicillin and EDTA (act on cell wall), SDS (act on cytoplasmic membrane), Bacitracin (acts on both cell wall and plasma membrane), Polymixin, Gramicidin S and Triton X-100 (triggering hypothetical lysine excretion channels). Polymixin gave the best results in terms of L-lysine HCL production. The L-lysine accumulated in the medium increased by more than 20 percent in comparison to that in control. The present study was the derivation of mutants using polymixin with/without nitrosoguanidine (NG). A polymixin sensitive mutant was found to increase the lysine accumulated in the fermentation medium by more than 10 percent by comparison to the parent.
