A method for detecting cell proliferation promoting factor in insects: Assay system using mitosis and DNA synthesis in the wing disc cells of Bombyx mori

We offer a system for detecting factors promoting cell proliferation in insects. Mitosis and DNA synthesis of Bombyx wing disc cells from the last larval instar were used as markers. The stage of wing discs, the addition of 20E and insect hemolymph were examined. Differences in the number of proliferating cells after culture in Grace (G), Grace containing 5 ng/ml 20E (GE) and Grace containing 5 ng/ml 20E and 10 % hemolymph (GEH) were observed in the V1 wing discs, while differences were not obvious in the V2 and V3 wing discs. Mitosis were observed in discs cultured in the Grace medium containing 5 or 10 ng/ml 20E. The addition of hemolymph increased the mitotic number in any 20E concentration. Mitosis resumed 18 hr after the addition of 20E and hemolymph, then it peaked at 36 hr. Among tested vertebrate growth factors, insulin revealed a promoting effect on wing disc mitosis and DNA synthesis after a 24 or 36 hr culture.
