Culture and regeneration from mesophyll protoplasts of garland chrysanthemum (Chrysanthemum coronarium L. var. spatiosum Bailey)

Culture methods for cell division, callus greening and regeneration of Chrysanthemum coronarium cv. sangoku-ithi mesophyll protoplasts were established. Characteristic of suitable culture medium for Chrysanthemum coronarium and other Asteraceae were the same. Ascorbic acid in additional medium for cell division suppressed browning reaction of cells. Zeatin was best cytokinin for suppressing browning reactions in regeneration culture, but zeatin was able to keep only 6.7% green calluses after 90 days of culturing. Subculture of only green calluses could be regenerated a bud, but had not been regenerated a whole plantlet
