Studies on some isozyme and glycoprotein patterns of maize seedlings grown in high salt concentration media

Aim of this research is the study on the optential of growth on high concentration salt media of high oil hybrid maize variety (KOSX 3503) which can grow well in the acidic pH media (Harinasut, P. et al., Full Research Report, KURDI, 1995). We use high oil hybrid maize variety (KOSX 3503), comparing with normal oil hybrid maize variety (KTX 3501) which come from inbred line Ki31 and their common parental varieties (Ki 32, Ki 42) as plant material. The callus was developed from seedlings of all maize varieties. Then, we study the potential of growth of all callus on agar media containing high salt concentration (NaCl 100 mM). We found that although all callus of studied maize varieties can physically well grow on agar media containing high salt concentration (NaCl 100 mM) within 4 weeks, the difference in development of some isozymes and glycoprotein can be detectable. These are the decreasing of peroxidase esterase and acid phosphatase activities and decreasing of glucose-mannose glycoprotein content of Ki 31 and Ki 32 when their callus were grown on agar media containing 100 mM NaCl or including being acidic pH of 5.5. However, the inbred line (Ki 42) and the hybrid varieties (KOSX 3503, KTX 3501) possess the more potential stress tolerance. Their activity of esterase and the glucose-mannose glycoprotein content do not decrease eventhough the callus culture media contain NaCl 100 mM or together with acidic pH of 5.5 within 4 weeks.
