Determination of Bt Toxin gene in transgenic tobacco plant for insect resistance by PCR analysis

Some preliminary results have been achieved in genetic transformation of tobacco variety K326 using Agrobaeterium tumefaciens on leaf disks. Agrobac terium tumefaciens strain EHA 105 with plasmid pITB2 containing bar, gus A and Bt genes was co-cultivated with segments of leaves. PPT resistant shoots have been obtained and they also expressed GUS activity. The presence of Bt gene was checked by PCR, a method for rapid application of specific target segments of DNA. DNA was extracted from the transformants and analyzed. The amplified fragments of Bt gene resulting from PCR have been determinated by electrophoresis on agarose gel
