[Effect of culture media on the regeneration of shallot ovaries (Allium cepa var. ascalonicum and A. x wakegi) grown in vitro]

Ovaries of shallot (Allium cepa var. ascalonicum) and wakegi onion (A. x wakegi) were induced for regeneration. The media used for culturing ovaries in this research were MS and B5 supplemented with 2 mg 2,4-D 1f at the power of -1 + 4 mg BA 1t at the power of -1. The stages of megaspore were 1 to 4,5 to 8 and 9 to 12 days before anthesis and cultured by Dunstan and Short (BDS medium). Analysis of the time interval needed for regeneration varied from 42 to 140 days after inoculation. The average induction rates of shallot and wakegi onion in B5 medium were 5.11 percent and 0.66 percent, respectively, while those in MS medium were 0.67 percent and 0.23 percent, respectively. The low percentage of regeneration induced by MS medium suggested that this medium is seemingly not suitable to promote embryogenesis of ovaries in Allium species. In the present study, the highest regeneration rate was 12 percent obtained by shallot ovaries harvested in one to four days before anthesis in B5 induction medium.
