Growth curve of Suwan 5 corn (zea mays L.) suspension culture

Embryos culture of Suwan 5 were culture on N6 medium supplemented with 2 mg/l 2,4-D, 1 mg/l NAA, 1 mg/l kinetin, 1 mg/l BA, 0.69 g/l L-proline, 100 mg/l casein hydrolysate, and 20 g/l sucrose which could induce numerous calli. Selected friable callus was transferred into liquid N6 medium and subculture period 10-15 days for 4 months, cell suspension culture was homogeneous and rapid growth. The growth phases of cell suspension at 0, 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 and 22 days respectively, were determined by fresh weight and dry weight measurement. The results showed that lag phase was at 0-4 days, log phase at 4-14 days after 14 days were in stationary phase and death phase. From this experiment, the transfer period was optimal at 4-14 days and it appeared to be useful for protoplast isolation subculture and regeneration.
