Allelic analysis of immunodominant major piroplasm surface protein genes of Theileria parasites in Thai cattle

Benign Theileria parasites DNAs of dairy and beef cattle from distinct geographical areas of Thailand were characterized by using PCR amplification with five sets of oligonucleotide primers. Two sets of them were modified from two genes of immunodominant major piroplasm surface protein (MPSP) coding for p32 of T. sergenti and p33/34 of T. buffeli. The other three sets of primers were basically generated from three alleles of MPSP which were specific for Japanese Theileria sergenti-Ikeda stock (I-type), Japanese T. sergenti-Chitose stock(C-type) and Australian T. buffeli-Warwick stock (B1-type), respectively. By using the allele-specific PCR, we found that the majority of Theileria parasites in Thailand was C-type MPSP whereas the mixed population of these types were minute. However, four groups of C-type MPSP were categorized when further PCR characterization of parasites using p32 and p33/34 features were achieved. The first group is presumed to be original Thai Theileria isolate (C-type+, p32 -, p33/34 -). The second group is similar to T. sergenti Chitose stock (C-type+, p32 -, p33/34 -). The third group is related to field isolate of T. buffeli from Australian cattle (C-type+, B1 -type +, p32+ and p33/34+). The last group is not to be thought of any groups of parasites (C-type+, p32+ and p33/34+). These results indicated that T. sergenti and T. buffeli have been distributed in Thailand.
