Improvement of Lentinus edodes (Berkeley) Singer BIOTECH 3084 by NTG mutagenesis for protein enrichment of corn cobs

Six putative mutants, namely: 3021, 4552, 4561, 6051, 6061, and 12031, were obtained from NTG mutagenesis of Lentinus edodes BIOTECH 3084. These were tested for their ability to increase the bioconversion of carbohydrate and degradation of lignin on corn cobs. Mutant 3021 yielded the highest soluble protein at 4.84 mg/g dry basis after 3 days fermentation. This mutant together with 12031 also showed the highest xylanase activity at 1.44 and 1.49 IU respectively, after 3 days fermentation. For ligninase activity, mutant 3021 showed the highest lignin peroxidase activity using the Azure B LP assay; however, it has negative phenol oxidase activity using the Bavendamm test. Among the mutants obtained, mutant 3021 is the most promising for protein enrichment of corn cobs. Stability of these mutants was also evaluated after five successive subculturing and no changes in colony growth in PDA plate and mycelial morphology were observed
