Isolation and identification of protease-producing bacteria from Mt. Makiling, Laguna [Philippines]

Three bacterial isolates (M sub 4-11, M sub 3-8, M sub 3-46) were obtained from soil and mud samples of Laguna. All of these isolates could grow at 45 deg C in nutrient agar and at 55 deg C if casein is present in the nutrient agar medium. Protease activity, observed as clearing zones in casein-containing plates, were noted for all bacteria at 37, 45, and 55 deg C. The results of the conventional identification techniques confirmed the initial identification of the bacterial isolates based on DNA sequence analysis of the 16S rRNA gene. All bacterial isolates were very closely related to Bacillus subtilis. One bacterium, M sub4-11, was identified as Bacillus subtilis and two were identified as B. pumilus
