Induced callus formation in chickpea "Cicer arietinum"

Research was conducted on Callus formation in chickpea Cicer arietinum varieties DG-89 and DG-92 at Sindh Agriculture University, Tandojam during 1998-99. Callus formation was observed in plants grown in pots in green house. After 7-15 days from growing seedlings explant material i.e., shoot tips, axillary buds, leaflets cuttings, and stem cuttings were excised, and cultured under laminar flow cabinet after sterilizing in sodium bychlorite solutions. The laminar flow cabinet along with other instruments was also sterilized in 70% alcohol and 80% spirit. Explants sliced in to pieces were transferred in to bottles containing different hormone levels. Data was collected for two weeks. After growing/formation of callus it was sub-cultured on different levels of hormones. Results showed that all explants indicated that callus could be induced in different utilized concentrations and combined BAP + 2, 4-D concentration. Variety DG-92 gave the best results.
