Milk protein characterization in domestic buffaloes and cattle using modified electrophoretic methods

Whole milk samples from 13 domestic buffaloes at the Philippine Carabao Center, UP Los Banos were examined to characterize their protein banding patterns using polyacrylamide gel electrophoresis (PAGE) and sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Both techniques revealed clear resolution of characteristics banding patterns and polymorphisms among and between buffaloes and cattle. PAGE distinguised buffaloes from cattle, while SDS-PAGE revealed variations among and within buffaloes and cattle. Using molecular weight standard, the study showed that both techniques resolve milk proteins of high molecular weights approximately between 50-100 kDa or over. This also suggests that among major milk proteins, albumins and immunoglobulins can be clearly resolved in PAGE and in SDS-PAGE gel. Further improvement in the conditions of both techniques is recommended to produce more superior banding resolution, and such proteins can be used as genetic markers for further genetic studies of these animals
