Evaluation of the genetic distance of sunflower genotypes (Helianthus annuus L.) using RAPD markers

14 genotypes of sunflower were screened for polymorphic RAPD markers with 13 RAPD primers. Strong PCR products were produced by 12 of these primers. 11 primers (91.7%) yielded polymorphic RAPD fragments. 1 primer was monomorphic. Of the total of 52 markers, 35 (67.3%) were polymorphic. The other 17 (32.7%) were monomorphic. The number of polymorphic fragments for each primer varied from 1 to 6, with an average of 3.2 fragments per primer. An average of 66.7% polymorphism per primer was revealed. The size of the amplified fragments ranged from 400 bp to 2000 bp. All the markers were scored by presence or absence of a specific amplification product. The genetic similarity values were calculated using Simple Matching (SM) coefficient and Jaccard's (J) coefficient. The Genetic Distance (GD) between genotypes were calculated as: 1 - SM and 1 -J. The values of genetic distances (Simple Matching coefficient) were between 21% and 58%, with an average of 38.5%, and between 31% and 78%, with an average of 51.2% (Jaccard's coefficient). The unweighted pair group method using arithmetic averages (UPGMA) was used to construct dendrograms.
