Optimation of endochitinase production from mycoparasitic fungi Trichoderma reesei

The objective of the experiment was to select optimal medium composition to induce endochitinase production of Trichoderma reesei. Growing media were prepared by substitution of 0.5 percent colloidal chitin, 0.5 percent colloidal chitin and 1 percent Polyvinyl Pyrrolidon (PVP), 1 percent crab shell chitin (Sigma), 1 percent crab shell chitin and 1 percent PVP into Richard's Medium (RM). Selected bioagent T. reesei isolate 13 was grown onto the media and the specific activity of endochitinase was measured. The results showed that T. reesei grown in all media performed enzyme expression. The highest specific activity of endochitinase was detected on T. reesei grown in RM medium substituted with colloidal chitin and PVP, followed by the same fungi in RM medium substituted with crab shell chitin plus PVP, colloidal chitin and crab shell chitin alone./Penelitian ini bertujuan untuk mendapatkan komposisi medium pertumbuhan yang paling optimal dalam menginduksi produksi endoktinase dari Trichoderma reesei. Medium pertumbuhan disiapkan dengan mensubstitusi sumber karbon, yaitu 0,5 persen koloidal kitin, 0,5 persen koloidal kitin + 1 persen Polyvinyl Pyrrolidon (PVP), 1 persen crab shell chitin (Sigma) dan 1 persen crab shell chitin + 1 persen PVP ke dalam medium Richard (MR). T. reesei isolat T13 ditumbuhkan ke dalam medium induksi tersebut dan diukur aktivitas spesifik endoktinasenya. Hasil penelitian menunjukkan T. reesei yang ditumbuhkan pada keempat medium induksi di atas mampu mengekspresikan endoktinase. Aktivitas spesifik endoktinase yang paling tinggi terdeteksi pada T. reesei yang ditumbuhkan pada MR + 0,5 persen koloidal kitin + PVP, diikuti MR + 0,5 persen koloidal kitin, MR + 1 persen crab shell chitin + 1 persen PVP dan MR + 1 persen crab shell chitin.
