[Production of protease produced by Bacillus megaterium DSM 319 and MS 961 using tofu liquid waste media]

Protease plays an important role in the industry as it is abundantly used by the food and non-food industries. Many studies have been carried out to find a cheap and readily available substrate for protease production. This paper reports the physical and chemical optimization for the growth of Bacillus megaterium DSM 319 and MS 961 in liquid tofu waste. The optimum condition for protease production using B. megaterium DSM 319 (max. activity 3.213 U/ml. min) was: concentration of tofu liquid waste 50 percent, pH 7.0 temp. 37 deg. C and agitation 250 rpm. In B. megaterium MS 961, a max. protease activity of 4.321 U/ml.min was produced with an optimum condition of: concentration of tofu liquid waste 25 percent, pH 8, temp. 37 deg. C and agitation 250 rpm. Ultration could improve protease activity in B. megaterium DSM 319 from 2.6 U/ml.min to 3.5 U/ml.min in concentrated volume 225 ml, while for B. megaterium MS 961 increase from 5.0 U/ml.min to 7.5U/ml.min in concentrated volume of 150 ml. However, protease activity produced in liquid tofu waste was still lower than those formed in the standard media G&C
