Development of inbred varieties: use of classical and DNA marker-aided backcrossing to transfer important genes or traits

The two BLB [bacterial leaf blight] resistance genes from pyramided lines (AR32-19-3-3 and AR32-19-3-4 with Xa21, Xa5 and Xa4 genes) were kept in IR64 and another in PSB Rc18 genetic background, but with a stemborer resistance gene from wild rice Oryza longistaminata. Another two BLB genes from pyramided lines were also kept in IR64 and BPI Ri-10 genetic background, but with a tungro resistance gene from Matatag 3, 6, IR73885-1-4-3-2-1-6, and other sources. Preliminary backcrossing resulted in 36 lines, incorporating tungro and stemborer resistance, and earliness with elite lines containing Xa-21 and Xa5. F2-derived lines are for further testing while another 65 entries are for ADN advancement. In the segregating population, 1518 entries were advanced to the next generation, and 4028 were selected during WS [wet season] for the 2004 trial. Eight elite lines contained Xa-21 and xa-5 resistance genes while two lines had either one of the genes. The new IR64 and PSB Rc14 were elevated to NCT [National Cooperative Test] phase 1, both having the xa-5 and Xa-21 genes known to be effective against 9 races of the BLB in the country. PR 31563-AR32-19-3-4 with IR64 background and advanced to NCT is now on its third season at MAT. Three elite lines with PSB Rc14 background and believed to contain restorer and BLB resistance genes were retained for further NCT testing
