In vitro induced colchiploids of citrus

Studies were conducted to explore the ploidy of important cultivars of citrus. Embryos of four cultivars were treated with 0.01 % and 0.10 % colchicine using in vitro conditions. The effect of colchicine on embryo germination, leaf characters and stomata was also observed. Mortality of embryos was wide spread with colchicine treatment in all the four cultivars. After colchicine treatment a significant increase was observed in leaf length and width. A general trend was observed that all cvs. showed significant differences for all the growth patterns and morphological studies. Microscopic studies of stomata showed a significant decrease in number of stomata in colchicine treated cultivars but length and width of stomata increased significantly. Root squashes were observed and it was found that the colchicine treatment yielded both diploid and tetraploid plants.
