Somatic embryogenesis and micropropagation of commercially important rice (Oryza sativa L.) varieties [Portugal]

Somatic embryogenesis and micropropagation systems were tested for some rice varieties. The varieties used in this study were Gladio, Thaibonnet, Koral and Basmati C620. A protocol was optimised for surface sterilization of mature rice embryos. Three culture media, differing in terms of nutrients and hormonal composition were compared for induction of somatic embryogenesis for each variety. Regeneration was obtained for all the varieties under study, although only Koral has already been transferred to soil. A micropropagation protocol was tested for the same four rice varieties. In optimal excision conditions, values of 8-15 shoots could develop from each inoculum. For Basmati C620, the best responding variety, up to 34 shoots/inoculum were obtained. A reduction in the sucrose content of the culture medium was also tested and found to improve regeneration. Micropropagated explants are currently being tested for rooting
