Callus induction and somatic embryogenesis from pistils culture of Citrus aurantifolia swing cv. Pantawai

Study on the effect of benzylaminopurine (BAP) on callus induction and somatic embryogenesis of lime (Citrus aurantifolia cv Pantawai) was conducted. Pistils were dissected from the flower buds before anthesis, and cultured on the Murashige and Tucker (MT) medium supplemented with 50 g/l sucrose, 500 mg/l malt extract and various concentrations of BAP for 12 weeks. The results showed that the MT medium containing 3.0 mg/l BAP gave the highest percentages of callus (83.3 percent) and somatic embryo formation (41.6 percent). Concentrations of BAP higher than 3.0 mg/l decreased the formation percentage. This micropropagation technique could be employed in the production of new varieties or disease-free citrus.
