Preparation and characterization of monoclonal antibodies against field isolated Aujeszky's virus in Thailand

Four monoclonal antibodies (MAbs) against field isolated Aujeszky's disease virus (ADV), namely 59D3, 85B8, 61E6 and 51D9, were produced and characterized. By Western blot analysis, MAb 59D3 and 85B8 recognized glycoprotein gII, while MAb 61E6 and 51D9 react with glycoprotein gIII. Using indirect immunofluorescence antibody (IFA) test, all MAbs were positive to the ADV used in MAb production, ADV Shope strain and 34 field-isolated ADV. By 50 percent plaque reduction nutralization test, MAbs 59D3 and 51D9 neutralized all the three groups of virus in the presence of the complement, whereas MAbs 85B8 and 61B6 neutralized the ADV used in MAbs production both in the presence and absence of the complement.
