Anther culture of hybrid rice (Khao Dawk Mali 105/Skybonnet)

Anthers of F1 hybrid rice (Khao Dawk Mali 105/Skybonnet) were cultured on the various callus induction media. It was found that a suitable medium for callus induction was N6 containing 2 mg/l 2,4-D,1 mg/l kinetin, 500 mg/l casein hydrolysate and 50 g/l sucrose. The plantlets could be produced from callus on the MS medium supplemented with 2 mg/l BAP, 1 mg/l kinetin, 0.2 mg/l NAA, 300 mg/l casein hydrolysate, 15 percent coconut water and 20 g/l sucrose. After transferring these plantlets onto the MS medium containing 1 g/l yeast extract, 15 percent coconut water, 3 mg/l BAP and 0.01 percent colchicine for 5 days, about 60 percent of the doubled haploid plants were recovered.
