Effect of temperature during centrifugation on the efficacy of nuclear polyhedrosis virus

A laboratory experiment was conducted to study the efficacy of Nuclear polyhedrosis virus purified at different temperature in deep freeze centrifuge and at different centrifugation speeds in ordinary centrifuge. The results showed that there was no significant difference between the POB's recovered at different centrifugation speeds viz., 2000 rpm to 10000 rpm though there was increase in temperature from 31.5øC to 38.5øC and there was no loss of virulence of Ha NPV. LT50 values varied from -5øC to 25øC. During centrifugation in deep freeze centrifuge, LT50 values varied from 117.53 h to 126.96 h. The rise in temperature in ordinary centrifuge during centrifugation (10000 rpm) did not have any deleterious effect on virulence of POB's which was below the critical temperature.
