EST library and normalized cDNA library as tools for probing the lupine - B. lupinii symbiosis

The cDNA library from yellow lupine roots 7 days after infection with B. lupinii (symbiotic bacteria fixing nitrogen) has been prepared and 480 cDNA clones were picked up randomly for sequencing and expression profiling. The EST library - the set of single sequencing reactions of each of cDNA clones - consists of 364 entries deposited in NCBI databases. The methods used for EST library analysis are presented. The EST library was organized into protein functional categories. Detail analysis of EST representing proline rich proteins reveales correspondence between the ESTs abundance and protein composition on cellular level. The same set of cDNA clones was used for expression profiling by means of macroarrays and reverse Northern. The modulation of gene expression was analysed in three organs: uninfected roots, roots infected with symbiotic bacteria and leaves
