Study of extraction method for the determination of Azadirachtin content during growth of neem (Azadirachta siamensis) Fruits

Method for extraction of azadirachtin by 95 percent ethanol and methanol has been investigated. It was demonstrated that the direct contact of grounded neem seeds kernel to solvent using magnetic stirrer at room temperature yielded higher extracted efficiency in comparison to method using Soxhlet apparatus at 120 deg C. The developing method exhibited over two folds quantity of azadirachtin recovery from neem seeds. This method was applied to monitor the variation in azadirachin content during growth development of neem fruits. Azadirachtin has been determined using reversed phase HPLC technique. Samples were collected during stage of flowers to ripened fruits. Result indicated that azadirachtin was appeared and first detected at the young flower stage. The level of azadirachtin was gradually increased and reaching the maximum at 12.5 percent dry wt. which in 7th week while the fruit color turn from green in to the deep-yellow. The amount of azadirachtin detected was sharply drop into 3.14 percent dry wt. in 9th week of fruit development which was equivalent to the deep-brown color of fruit surface.
