[Identification of gene influencing the carotenoid pigments production by Pantoea agglomerans bacteria]

Indentification of mutant gene of Pantoea agglomerans 1E bacteria characterized by increased production of carotenoid pigments was presented. The mutant gene was cloned and sequensed. The gene homology according to the aminoacids contents with the hnr (rssB) E. coli gene was found. RssB-albumen took part in degradation of RpoS – subunit of RNA E. coli polymerase which was necessary for transcription of some genes in stress conditions. Perhaps the regulation mechanisms of RpoS dependent genes expression were similar in E. coli and P. agglomerans. In course of the logarithmic phase of growth the RssB-albumen (product of rssB gene) interacted directly with RpoS. Further, this protein complex was recognized by ClpXP-protease which degraded the RpoS. In course of rssB gene mutation the amount of RpoS in P. agglomerans IE cells increased during the exponential growth phase. This leaded to early expression of RpoS-dependent genes including the genes responsible for carotenoid biosynthesis. That is why the mutant bacteria began to produce the pigments earlier and could accumulate the larger amounts of carotenoids in comparison to wild type cells
