Mass propagation, growth promotion and acclimation methods by tissue cultures for ananas

The present studies were carried out in order to establish the method of propagation, promotion of growth and acclimation by tissue culture for Ananas. Axillary buds of Ananas(Vriesea carinata) were used for culture. The primary culture, Multiple shoots were formed from the axillary buds on a basal medium(MS) supplemented with 2.0 mg/l Naphthalene Acetic Acid (NAA)and 2.0 mg/l Benziladenine (BA)and 30 g/l sucrose. The Secondary culture, Multiple shoots of the primary culture also grew up very well on the same basal medium supplemented with 2.0 mg/l BA and 30 g/l sucrose. Multiple shoots were divided and transplanted, they readily rooted on MS medium supplemented with 30 g/l sucrose without hormones. Rooting shoots were acclimated. The bog moss was suitable for the acclimation as the planting materials and the optimum temperature was 15-30 deg C. To the above-mentioned, it was possible to propagetion from one multiple shoot to 500 seedlings or more of this culture method for two years. However, the growth of the Ananas was very slowly. Therefore, it took 3-6 months from the division of the multiple shoots to rooting. Moreover, it took 6-12 months from rooting to the acclimation was required.
