Enhancing growth of roots of Yaa Phraek Hin (Ophiopogon intermedius) D. Don by microorganisms

Three strains of Agrobacterium rhizogenes namely ATCC 15834, A4 and R1000 were used to induce hairy root in Yaa Phraek Hin (Ophiopogon intemedius) D. Don, which is monocotyledonous plant, in family Liliaceae, the roots were used as active ingredient in some of herbal medicine, The results showed that Yaa Phraek Hin had no responed to hairy roots induction from all strains of Agrobacterium rhizogenes. Soil samples around roots of Yaa Phraek Hin were collected from natural forest. Twelve species of VAM fungi were isolated from soil and tentatively identified as Acaulospora sp. and Glomus spp. All spores of VAM fungi were propagated in corn. Only nine species could be propagated. These nine species and four species namely Acaulospora scrobiculata, Scutellospora calospora, Glomus occultum and scutellospora sp. No.1 from department of microbiology were inoculated to shoots cutting and plantlets of Yaa Phraek Hin which were micropropagated by tissue culture. The results showed that four species namely Glomus sp. No.18, Glomus sp. No.22, Scutellospora sp. No.1 and Glomus occultum had potential to promote growth of Yaa Phraek Hin. The experiment was also made on effect of those four species of VAM fungi on the growth of Yaa Praek Hin. Only Scutellospora sp. No.1 was proved to enhance the growth of Yaa Preak Hin. Growth parameters such as fresh weight and dry weight of roots and leaves, total fresh weight and total dry weight of roots and leaves, total fresh weight and total dry weight of roots and leaves of Yaa Phraek Hin from treatment inoculated with Scutellospora sp. No.1 were highly significant higher than control.
