Strain status of Pplum pox virus in Bulgaria

Samples from Prunus hosts, exhibiting typical Plum pox virus (PPV) symptoms were collected from orchards in Bulgaria and studied by DASI-ELISA test and IC-RT-PCR analysis with strain specific primers, targeting coat protein (CP) and cylindrical inclusion protein (CI) regions for determination of their strain status. Using DASI-ELISA test 45 samples were proved to be PPV infected. The results obtained showed that 31% of the isolates belonged to PPV-M strain, followed by PPV-D strain (28%) and PPV-Rec strain (12%). Mix infections of M+Rec (4%) and M+D(4%) strains were found, too. Only -M strain was identified on peach from all region tested, while both strains, -M and -D were identified on plum infected trees. A good correlation between the results of DASI-ELISA test with the use of strain-specific monoclonal antibodies (MAbs) and PCR analysis with the use of strain-specific primers was noted.
