Differential gene expression in somaclonal individuals of sugarcane resistant to fungal diseases have been studied by AFLP-Cdna. Somaclonal genotypes were selected in field trial for their resistant to rust (Puccinia melanocephala) from variety B4362; to smut (Ustilago scitaminea) from either Ja6O-5 or C85-101 and to eyespot (Drechslera sacchari) from C87-51, respectively. Procedures for AFLP-Cdna were made following the instructions reporter by Bachem y col. ww.keygene.com). In all cases, transcript derived fragments (TDFs) were found to be differentially expressed between somaclonals and the donor variety. For smut, a total of 52 Cdna fragments were evidenced polymorphics associated with one genotype. For rust, a total of 70 polymorphic Cdna fragments were found differentially expressed as following: 34 with one genotype, 16 with two genotypes, 18 with three genotypes and, 2 with four genotypes. In the case of eyespot 18 TDFs were polymorphics: 16 associated with one genotype and 2 with two genotypes. Sequencing of polymorphic TDFs is under way. Implications for the functional genomic programm in sugarcane are discussed.
