Sugarcane is a major crop in Brazil being cultivated in more than 5 million hectares, from the south to the northeast. Its importance for sugar and alcohol production justified the first plant genome program by the ONSA network founded by FAPESP in São Paulo State. Today, more than 259,325 reads (around 40,000 clusters) were sequenced and deposited in the Sugarcane ESTs Project (SUCEST) database. More than 200 people are working in this project lacking for genes involved in different pathways. Our group is investigating those genes involved in plant resistance by the localization of bacterium at the invasion site and avoidance of its spread through the vascular system. Each mechanism is presented and discussed. Also, gene products related to each defense mechanism with homology with sugarcane ESTs were identified and annotated from the SUCEST database. Several sugarcane ESTs homologous to genes related to pathways involved in phytoalexin synthesis, tyloses formation, calluses deposition, phenolic and gels infusion and lignifícations of, tissues were identified. Even preliminary, the results open an avenue for studies directed to the identification of all factors used by plants to defend themselves against pathogenic bacteria inhabitant of xylem vessels. Once challenged by such pathogens, the pattern of gene expression during pathogenesis in the xylem vessels could be easily assessed by the DNA chip technology.
