Development of an in-vitro culture protocol for plantlet regeneration in squash (Cucurbita moschata Duch.) cv. Rosalinda

Shoots were regenerated in-vitro from cotyledons of ungerminated seeds after culture for 3-4 weeks in MS basal medium supplemented with BA, with or without auxin. Regenerated shoots readily formed roots on MS basal medium and grew to normal plants upon transfer to soil. Cotyledon, hypocotyl and root explants taken from in-vitro grown seedlings formed only callus and/or roots when cultured in MS medium supplemented with auxin (IAA or 2,4-D) and cytokinin (BA or Ki). All cultures died within four weeks of incubation. Attempts to establish cultures of epidermal peels of the cotyledon and of a proliferating cell suspension culture had not been successful. The present study is the first successful attempt on plantlet regeneration via organogenesis in C. moschata Duch. cv. Rosalinda. The protocol presented may be harnessed for genetic transformation studies in local squash.
