The soil, as a significant link in the chain of matter and energy cycling in the field, is very often subject to contamination derived from radiation accidents and local radioactive sources. In the aim of timely identification and undertaking the adequate protection measures, it is necessary to study the soil investigation methodology in the case of radioactive contamination. This paper presents the methods of soil investigation in the case of radioactive accident, as well as the method of soil investigation under local contamination by radioactive substances. In the case of an accident, we present the soil sampling procedures of cultivated soil, procedure of non-cultivated soil and the procedure of soil sampling together with the plants. In the case of local contamination by radioactive substances, we present the criteria of identification and classification of the sites with local radioactive contamination in agriculture. In addition to the soil and plant sampling techniques, we also present the methods of sampling the products of plant and animal origin. The presented methods of the research of radionuclide migrations in agro-ecosystems include the study methods of air pollution of land - plant cover and the study methods of lateral and vertical migrations of radionuclide in the soil. Aiming at the maximally reliable study results, we also present the assessment method of data reliability and the stochastic characteristics of radionuclide inputs in agricultural crops. Taking into account the soil heterogeneity as the three-phase system, the lysimetric investigations in the research of radionuclide migrations in agro-ecosystems is also presented. The paper presents the methods applied for the determination of radionuclide contents, as well as the safety technique in the handling of radioactive substances.
