Design and construction of fermentor for enzyme production from Humicola lanuginosa

Raw material used in this experiment was agricultural waste that the ratio of corncob : wheat bran : rice husk was 5 : 5 : 2. The initial moisture content was 70 percent. Humicola lanuginosa was cultivated at 45 deg C in 250 ml erlenmeyer flask and 5 liter cylindrical fermentor. The results show that 4 ml/12 g dry solid spore suspension concentration is good ratio for this cultivation. Shaking flask by hand gives lower enzyme activity than the static flask and shaking flask in the temperature control water bath. The investigation in the 5 liter cylindrical fermentor show that the system is couple of aeration with ambient air and moist air are better than the non-aeration system resulting the higher enzyme activity. The maximum enzyme activity of 1,047.12 units/g dry soil is observed when the moist air is fed at 0.12 l air/g dry soild/h. The effect of bioreactor rotation on the enzyme activity was also investigated. The start up rotation at 24 h and 48 h gives the same enzyme activity of time of 1,047.12 units/g dry solid at different batch time of 96 and 72 h. Therefore, the start up time of 48 h was recommended to save the operating cost and fermentation time. Finally, the different substrate loading was varied to 550, 825, 1,100 g which obtain the enzyme activity of 1,047.12, 976.34 and 890.05 units/g dry solid.
