The research has been conducted in brackishwater pond area in Lengke I and II, Bambu and Simboro Mamuju Regency, South Sulawesi. The aim of this research was to study the differences of individual number, diversity indices, dominance indices, and evenness indices and relationship between substrate (organic matter, sand, clay and silt contents) and the diversity of macrobenthos that were settle in brackishwater pond soil. Sampling to obtain macrobenthos in the soil conducted through grabbing the soil in the plots each size of 25 x 25 x 10 cubic cm representing the pond area closes to the road, the pond in transition area, the area closes to the beach. Results of this research showed that the diversity indices were significantly different between Lengke II with Lengke I, Bambu, and Simboro (P less than 0.05). Dominance indices were significantly different (P less than 0.05) between Lengke II with Lengke I while individual number and evenness indices of macrobenthos were not significantly different (P more than 0.05). The organic matter content appears to have a stronger relationship (R at the power of 2 = 0.3193) with the diversity of macrobenthos compared with the dominance of macrobenthos (R at the power of 2 = 0.1833). The clay tends to have a stronger relationship (R at the power of 2 = 0.3093) with the diversity of macrobenthos compared with the dominance of macrobenthos (R at the power of = 0.2234)
