In vitro selection for aluminium tolerance on somaclon variants of two tomato cultivars (Lycopersicon esculentum Mill.)

The objectives of the research were to obtain suitable combination on basic medium and growth regulators for inducing callus formation and organogenesis in tomato callus, and to conduct in vitro selection for Aluminium tolerance on somaclon variants of two tomato cultivars, San Marino' and 'Ratna'. The research was divided into two experiments, both of which were arranged in a completely randomized design (CRD). The first experiment resulted that MSO with IAA 1 mg/l + Kinetin 2 mg/l was the most suitable and efficient medium to induce callus formation and organogenesis on tomato. The second experiment showed that callus of San Marino and Ratna had the same level of Al tolerance. The highest level of Al in which the callus could regenerate shoots was 1000 micro M Al. The shoots selected in medium with 1000 micro M Al succeeded to develop roots in the rooting medium of MSO + IAA 0.1 mg/l with up to 1500 micro M Al. There were three plantlets obtained tolerance to 1500 micro M Al. One from 'San Merino' and two from 'Ratna'.
