Study on relative resistance of local cultivars of alfalfa to Alfalfa Rust Disease

Abstract Since alfalfa rust disease can damage to alfalfa yield specially in seed production fields, to evaluating relative resistance of cultivars to this disease 10 cultivars were used in RCBD trial with four replications in Gorgan (field test) and Karaj (green house and laboratory tests) regions during 2002 and 2003. Infection of cultivars in natural conditions were determined based on the number and size of pustules on the leaves in nine level, 1 to 9, in early summer in field trial (Hill et al, 1963). In green house trial, 20 days-old seedlings of alfalfa were inoculated by spraying of suspension of Uromyces striatus Schroet spores containing 3.5 پ~ and#1633and#1632and#1637spore per milliliter in humid chamber .The inoculated seedlings were incubated in standard conditions and Infection Types of cultivars were recorded based on a scale containing 0 to 4 levels (Koepper, 1942). In laboratory test, the Excised Leaflet Inoculation assay was used. T he inoculated leaflets in petridishes containing %3 sucrose solution were incubated in a germinator with 21oC under fleurecent light for two weeks and then Infection Types were determined based on a scale containing 1 to 5 classes (Hill et al., 1963). The results of field evaluation and the analysis of variance of data showed that difference between treatments were statistically significant (ƒ؟=%1) in responding to disease and totally the most susceptible cultivars were Renger and Hamedani and the most resistance cultivar was Malekkandi and other cultivars showed relative tolerance to disease. The results of green house and laboratory evaluations showed that Hamedani and Renger cultivars had relative susceptibillity to disease and Mohajeran and Rahnani cultivars had more relative resistance to disease with comparison to other cultivars.
