Effect of medium type and incubation duration on improved in vitro tuberization in three Dioscorea rotundata Poir cultivars

The effect of sequential treatment (6, 8 or 10 weeks) with tuberization medium (T-medium) followed by Murashige and Skoog medium (MS medium) on the sizes and weights of microtubers in shoot cultures of white yam (Dioscorea rotundata Poir) was evaluated. Three local Ghanaian white yam cultivars (Puna, Punjo and Kablete) were used. The treatments were found to have substantial effects on microtuber induction in the cultivars studied. Culturing the explants on T-medium for 8 and 10 weeks followed by MS medium (full strength) gave the highest microtuber induction, frequency and yield and highest individual microtuber weights. Microtuber fresh weight:dry weight ratios were higher for both the 8 and 10 weeks transfer treatments than the 6 weeks transfer and the control. Cultivars left on the T-medium for 6 and 10 weeks yielded higher shoot numbers than the control and the 8 week treatment
