Presence of bovine herpesvirus 4 (BHV-4) in population of cattle and methods of laboratory diagnosis: [Master thesis]

Presence of BHV-4 infection in most European countries, initiate searching for presence of BHV-4 in Serbia and also considering effects of BHV-4 infection on health of heifers and cows with intensive milk production in our country. Investigation of presence of BHV-4 infection was performed on two farms of dairy cows near Belgrade with a high rate of postpartum metritis. In order to complete epizootical view, serological examination was also done on cows in individual sector. Serological test was implied on 240 samples from two farms near Belgrade and 51 mini farms in Belgrade epizootical area. The percent of serological positive cows was 84.50% on farms and 3.75% on mini farms. Virological testing was done using vaginal content of cows, from 3 to 30 days after partus, and the result was isolation of three BHV-4 isolates. After use of PCR, specific parts of genome BHV-4 (MDBP, IE2, TK) were amplified. Using restriction analysis, with BamHI and EcoRI endonucleases, genotypization of isolates and comparison with referent European and American referent strains was performed. Our isolates are very similar to European group BHV-4 viruses - MOVAR. This was the first isolation and classification of BHV-4 isolates in Serbia.
