Influence of pretreatment on hop cryopreservation

In our experiments we compared different pretreatment on hop (Humulus lupulus, L.) cryopreservation. We evaluated impact of 0.75 M sucrose and 60% polyethylene glycol together with low temperature treatment on survival and regeneration of hop plant after cryopreservation. We found out that pretreatment by both osmotic components had significant effect on an improvement of survival and plant regeneration. The highest plant regeneration was obtained for the method using nodal cuttings pretreatment with 0.75 M sucrose and low temperature treatment (2 C) and following shoot tip loading with 0.7 M sucrose for overnight and air desiccation of shoot tips above silicagel (3 hours). This method is used routinely for cryopreservation of hop germplasm at the Research Institute of Crop Production in Prague in collaboration with the Hop Research Institute, Co., Ltd. in Zatec.
