Deep frozen semen quality of bull processed with whole and fractionated dilutions of tris yolk and tris yolk-raffinose

Deep frozen semen quality of 12 bulls aged 4-5 years were compared using two diluters, tris yolk (TY) and tris yolk-raffinose (TYR), processed with two methods, whole and fractionated dilution methods Motility of spermatozoa before freezing were not significantly different, similar to the motility after thawing at 0 hour (P GT 0.05). Stress test at 37+-1 deg C for 5 hours period after thawing revealed that at 2 and 3 hours spermatozoa in semen diluted with the whole and the fractionated dilutions of TYR were significantly tolerable than spermatozoa diluted with the fractionated dilutions of TY. The motility at 2 hours were 42.3 percent and 41.5 percent compared with 37.5 percent (P LT 0.05) the motility at 3 hours were 28.4 percent and 28.8 percent compared with 22.0 percent (P LT 0.05) respectively. However, they were not significantly higher than the method using whole dilution of TY (P GT 0.05). Sperm motility in semen processed with the whole and the fractionated dilution methods were not different. The whole dilution method and the TY dilution are more economic and convenient for routine semen production of the A.I. Centre.
