Relationships between endogenous hormone contents and cytoplasmic male sterility in sugarbeet

Chino. 研究甜菜细胞质雄性不育与内源激素含量的关系。以表现型具明显差异的两对甜菜单胚CMS系及其O型系作为试验材料,采用间接酶联免疫(ELISA)检测技术,分析营养生长期和生殖生长期的功能叶片和花蕾中生长素、细胞分裂素、脱落酸、赤霉素等4大类内源激素的含量和比值变化。在营养生长的糖分积累期,ABA、IAA、GA3含量均为O型系高于CMS系,差异显著,而DHZR、IPA、ZR含量在营养生长期间均差异不显著;在开花初期的叶片,除IPA外,IAA、ABA、DHZR、ZR、GA3含量均为O型系高于CMS系;饱满花蕾的IAA、ABA、ZR、GA3含量均为O型系高于CMS系。甜菜激素变化较有规律的时期和器官依次为开花盛期的饱满花蕾、开花初期的叶片和和营养生长阶段糖分积累期的叶片。在上述3个时期, CMS系与O型系甜菜的激素含量变化较大,CMS系的IAA/ABA、ZR/ABA和GA3/ABA值分别为O型系的0.55倍、1.43倍、1.64倍;2.66倍、1.55倍、1.49倍和1.14倍、1.60倍、1.53倍。各种激素的含量变化与甜菜CMS相关密切。影响甜菜CMS的时期、器官和激素比值分别为饱满花蕾IAA/ABA的降低、ZR/ABA和GA3/ABA的提高,开花初期和糖分积累期的叶片为IAA/ABA、ZR/ABA和GA3/ABA的提高。该研究为探讨甜菜细胞质雄性不育的生理机制和育性的调控提供了理论依据。
Mostrar más [+] Menos [-]Inglés. This study was designed to elucidate the relationship between endogenous hormone contents and cytoplasmic male sterility of sugarbeet. Enzyme-linked immunosorbent assays (ELISA) was used to determine indoleacetic acid (IAA), cytokinin, abscisic acid (ABA) and gibberellin (GA3) contents and the ratio of these endogenous hormones to ABA, respectively. These endogenous hormones were taken from functional leaves and buds of two pair of monogerm cytoplasmic male sterile (CMS) lines with significant different phenotype and O-type line sugarbeet during vegetative and reproductive period. ABA, IAA and GA3 contents in leaves of O-type line were significantly higher than those of CMS lines, except DHZR, IPA, ZR, during vegetative period, to the early stage of flower, IAA, ABA, DHZR, ZR, GA3 contents of CMS lines were higher than those of O-type line, except IPA; while in buds IAA, ABA, ZR and GA3 contents of CMS lines were lower than those of O-type line. The regular changes of theses endogenous hormones contents in organs were found in leaves during vegetative period and at early stage of flower, and in buds during florescence. For the three periods, compared with O-type line, the ratios of IAA, ZR and GA3 to ABA for CMS lines were 0.55, 1.43, 1.64; 2.66, 1.55, 1.49; and 1.14, 1.60, 1.53 times, respectively. The changes of endogenous hormones contents and CMS are closely connected. Decrease in ratio of IAA to ABA, while increase in ratios of ZR and GA3 to ABA in bud, and increase in ratios of IAA, ZR and GA3 to ABA in leaves during vegetative period and early stage of flower were important factors involved in CMS. This study has provided a fundamental basis for further research on physiological mechanism and regulation of cytoplasmic male sterility in sugarbeet.
