Variavilidad molecular de aislados de begomovirus que afectan los cultivos de pimiento en Cuba

The presence of begomoviruses in pepper crops was reported for first time in Cuba in 2002, with the detection of Tomato yellow leaf curl virus (TYLCV). Later, infection of peppers plants in the eastern region countries by a new bipartite begomovirus with 94% identity with Cabbage leaf curl virus (CaLCuV) was determined. To investigate the molecular diversity of these viruses, a survey was carried out in 2007 in the main areas for peppers production in Cuba. Pepper samples showing typical symptoms of viral diseases caused by begomoviruses were collected in number of 212. The collected samples were initially tested by PCR using begomovirus-universal primers (Palv1978/ PARc496), and 30 percent of them were positive for infection. DNA extracted from these samples was used as a template for full-length circular genome amplification using Rolling Circle Amplification (RCA). The amplified products were characterized by Restriction Fragment Length Polymorphism (RFLP), which allowed detecting of fours distinct patterns in the analyzed samples. This result indicates the possible presence of two additional viruses, besides the two previously reported begomovirus species, and constitutes evidence of the wide molecular variability of theses pathogens in Cuba. The identification of these viruses by sequence analysis will provide more detailed information about the molecular diversity of begomoviruses infecting peppers in Cuba.
