A convenient comparative method using a simple pretreatment process for comparison of quercetin glycoside contents in onions (Allium cepa)

We investigated a pretreatment method of spectroscopic analysis of samples for convenient evaluation of quercetin glycoside contents in onion varieties or breeding lines. We clarified the pretreatment process and processing time, and we showed that the sample preparation time and amount of reagent used can be reduced by approximately 90% and 95%, respectively, compared to those in the previously reported analytical protocol. The absorbance values, measured at 360 nm, of the extracts prepared by the procedure were highly correlated with the values of HPLC analysis (r=0.93), indicating that the simple evaluation method can be used for determining relative quercetin content of examined varieties. In addition, the pretreatment procedure was easy to carry out and had a high throughput. Thus, it was concluded that the simple evaluation method developed in this study is suitable for screening breeding lines, and it is expected to aid in the development of onion varieties with high quercetin glycoside contents.
