A simple spectrophotometric method for the estimation of beta-carotene content in spinach acetone extracts

A simple method for the estimation of beta-carotene concentration in acetone extracts of spinach was newly developed. The principle of this method depended on the difference in visible absorption spectra of chlorophylls and carotenoids. The concentration of beta-carotene can be obtained through the calculation with the absorbances at 479, 645 and 663 nm. The equation, the correlation coefficient between prediction and measurement (r) and the standard error (SE) are as follows. beta-Carotene (mg/L) = 0.854Asub(479) - 0.312Asub(645) + 0.039Asub(663) - 0.005 (r = 0.992, SE = 0.0096 mg/L). Only an ordinary spectrophotometer is necessary to use this method, and it is easy enough to be used for many samples.
