Genetic and breeding study on near-isogenic lines of IR64 for yield-related traits-selection of core sets for studies on adaptability to water-saving circulation

A total of 334 near-isogenic lines (NILS BC3- derived lines) derived from crosses between a recurrent parent IR64 and a donor parent of one of 10 high-yielding varieties (IR65600-87-2-2-3, IR65598-112-2, IR65564-2-2-3, IR66750-6-2-1, IR69093-41-2-3-2, IR69125-25-3-1-1, Chugoku 146, IR66215-44-2-3, IR68522-10-2-2, and IR71195-AC1) have been developed by back crosses breeding. Variations of these NILs in the genetic background of IR64 were evaluated in terms of agronomic traits, such as days to heading, leaf length, leaf width, culm length, panicle length, number of tillers, grain number, and grain weight. To determine introgressed chromosomal segments from the donor parents, more than 200 SSR markers covering the whole genome were used to analyze the graphical genotype of 334 NILs. Core sets representing variation of 334 NILs were selected based on phenotypic data collected in the 2005 wet season (WS) 2006 dry season (DS), 2006 WS, and 2007 DS. A total of 117 NILs, which were significantly different from IR64 in agronomic traits, were selected as a core set. Significant difference in traits were constantly observed in more than three seasons between IR64 and the selected NILs. Of the 117 NILs, 22, which had more than unique traits, were selected as a mini core set for detailed study. The selected lines also represented genetic diversity of 334 NILs as revealed by cluster analysis based on genotypic data. These selected lines were used in adaptability tests under water saving cultivation, where a smaller number of breeding lines could be tested. These core sets a useful tool to analyze unique agronomic traits and important breeding materials.
